    摘要:

    为探究水中悬浮物含量对斑马鱼(Danio rerio)胚胎发育的影响,挑选发育正常的斑马鱼胚胎分为CK组、T1组、T2组和T3组,分别在悬浮物质量浓度为0、10、100和1 000 mg·L -1的水体中培养168 h,每12 h记录胚胎孵化率和胚胎或仔鱼的死亡率,在实验进行72 h后统计仔鱼心率,在实验进行168 h后统计仔鱼畸形率,并于实验进行4、12、24和48 h后拍照观察胚胎的发育情况。结果显示:1) 与CK组相比,T1组、T2组和T3组斑马鱼胚胎的累计孵化率在实验进行60 h后有统计学意义上的下降(p<0.05),且T3组的该项指标在实验进行72 h后直至实验结束均有统计学意义上的下降(p<0.05);2) 在实验进行120 h后直至实验结束,T3组与CK组相比仔鱼的累计死亡率均有统计学意义上的上升(p<0.05);3) 在实验进行72 h后,T1组、T2组和T3组与CK组相比仔鱼心率有统计学意义上的下降(p<0.05);4) T3组与CK组相比仔鱼畸形率有统计学意义上的上升(p<0.05);5) 与CK组相比,T1组、T2组和T3组的胚胎发育较为迟缓。研究结果提示水体中悬浮物质量浓度过高可导致斑马鱼胚胎发育迟缓、胚胎孵化率和仔鱼心率下降、仔鱼的死亡率和畸形率上升。

    Abstract:

    For investigating the effects of suspended solids content in water on the development of zebrafish (Danio rerio) embryos, the normally developing embryos were selected and placed in water with suspended solids mass concentrations of 0, 10, 100, and 1 000 mg·L -1 for 168 hours (h), and were named CK group, T1 group, T2 group and T3 group, respectively. During this period, hatching rate of embryos and mortality of embryos or larvae were calculated every 12 h, the heart rate and deformity rate of larvae were recorded after 72 and 168 h of the treatment, and the development of 〖JP〗embryos were photographed after 4, 12,〖JP〗 24, and 48 h of the treatment. The results showed that: 1) Compared with the CK group, the cumulative hatching rate of embryos in T1 group, T2 group and T3 group decreased statistically after 60 h of the treatment, and this indicator was reduced obviously in the T3 group after 72 h of the treatment until the end of the experiment (p<0.05); 2) After 120 h of the treatment until the end of the experiment, the cumulative mortality rate of larvae in the T3 group had statistically increased compared with CK group (p<0.05); 3) There was a statistically significant decrease in heart rate of larva after 72 h of the experiment in groups T1, T2 and T3 compared with group CK (p<0.05); 4) Compared with the CK group, the deformity rate of larvae in the T3 group increased statistically (p<0.05); 5) Compared with the CK group, embryos in T1, T2 and T3 groups were delayed in development. The results indicate that the high mass concentration of suspended solids can lead to delayed development of zebrafish embryos, decreased hatching rate of embryos and heart rate of larvae, and increased mortality and deformity rates of larvae.
